UV cut off - HPLC #2

  Sensitivity  ของ  detector  พิจารณาในเทอมของ  signal/noise  เมื่อ  mobile phase  มีการดูดกลืนแสงมาก  ความ
เข้มแสงที่ปล่อยผ่าน  flow cell  ของ  detector  จะน้อยนั่นคือ  baseline noise  มีค่ามาก  สำหรับ  HPLC  ในทางปฏิบัติแล้ว
จะไม่เลือกใช้ความยาวคลื่นที่  background absorbance  ของ  mobile phase  อยู่ใกล้ค่า  UV cut-off  ซึ่งนอกจากจะทำให้
baseline noise  มีค่ามากแล้วยังทำให้  linear dynamic range  เปลี่ยนไปอันเป็นผลมาจาก  mobile phase  มีการดูดกลืนแสง
มากและ  background absorbance  มีค่าสูง

 ดังนั้น  การที่จะให้ค่า  sensitivity  สูงสุด  ผู้ใช้ควรเลือก mobile phase ที่มีค่า  UV cut-off  น้อย
กว่า  wavelength  ที่เลือกใช้ในการวิเคราะห์ตัวอย่าง   ตารางข้างล่างแสดงค่า  UV cut-off  ของ  mobile phase  ซึ่งจะช่วย
ในการเลือก  mobile phase  ที่เหมาะสมในการวิเคราะห์ทาง  HPLC  ที่ใช้  UV Detector
